Tissue Expression and Environmental Adaptability Analysis of Intermediate Filament Protein Gene of Onchidium reevesii

As one of the main components of cytoskeleton, filaggrin plays an important role in maintaining the normal cell morphology and the directional transport of intracellular substances. In order to explore whether filaggrin intermediates play a corresponding role in the sulphogenic deformation capacity and post-injury repair process, the full-length cDNA sequence of filaggrin intermediates was cloned by RACE method, and the bioinformatics analysis and rt-qrcr experiment were conducted. The results showed that the total length of the gene sequence was 1 957 bp, and the sequence analysis showed that the ORF frame of os-if1 was 1 359 bp, encoding 452 amino acids, with a length of 376bp at 5 'UTR and 220bp at 3' UTR. The theoretical molecular weight is 51.54KDa, and the isoelectric point is 5.21.Both the secondary and tertiary structure prediction results showed that there was a very conservative helical domain in the middle of the polypeptide chain, which fully conforms to the dumbbell-shaped configuration of the filagin family. Fluorescence quantitative results showed that although the gene was widely distributed in various tissues and organs, it also played a role in the repair of epidermal injury. Therefore, MrBayes and IQtree were used to construct the phylogenetic tree of or-if gene. Through the analysis of the full-length cDNA cloning and related functions of the filagentin gene, this paper hopes to provide basic data for the further study of the repair of the sulfonate injury of the tumor, and more importantly, to provide valuable molecular basis for the in-depth study of the mechanism of the adaptation of invertebrate skin tissues to the change from aquatic to terrestrial environment.
